1. 萬能抗體鎖研究 (鎖盡天下名刀): 讓抗體藥更具選擇性
抗體藥物已為現今臨床主流(2013年產值540億美金)藥物,但其所辨認的抗原亦於正常組織表現,長期使用常易產生副作用。因此如何再增加抗體選擇性有其必要性,我們的策略是利用抗體自身之Hinge domain結構作為抗體鎖(閉鎖器),以蛋白酶受質胜肽連結抗體和此閉鎖器,形成具屏蔽抗體結合能力之閉鎖器-抗體,唯有在蛋白酶過度表現的疾病區,閉鎖器才能被移除,回復抗體功能,增加抗體對疾病區域之選擇性,並降低全身性副作用,提供病人更好的生活品質。未來抗體鎖每鎖1抗體藥就可產生1技轉,可多次技轉給全球各大藥廠,預計其專利具有100億美元以上的商機。(目前進行中疾病抗體:類風濕性關節炎(Humira、Remicade)、骨質疏鬆症(Prolia)、動脈硬化 (anti-VCAM)、牛皮癬(Stelara、Raptiva)及癌症(Herceptin、Yervoy、Panitumumab、Tremelimumab)
圖一:可提升抗體藥物選擇性的萬能抗體鎖:抗體鎖可鎖盡天下所有名刀(抗體)的結合能力,唯有在疾病區,抗體鎖才能被移除,回復抗體功能,增加抗體對疾病治療之選擇性,並降低全身性副作用。假設,每一個抗體都是一把名刀,在我們的體內殺遍抗原,以治療疾病,但不是所有的抗原都是壞人,不能全殺無赦,因此就需要" 刀鎖"的存在。刀鎖就像刀鞘,使名刀(抗體)在正常組織藏匿刀鞘裡不露鋒芒,唯有到達疾病區域時才會出鞘,砍殺壞抗原,降低名刀全身性鋒芒畢露的致命缺點(副作用)。本刀鎖可鎖盡天下名刀,讓其靈性(選擇性)升級,未來適用於提升全球百大藥廠的名刀,增加其疾病治療時的靈性,因此商機無限!
2. 電腦結構輔助抗體人類化研究 (電腦做抗體):抗體人類化與最佳化
抗體是人體最天然的藥物,所謂"天然的最好"抗體新藥已成為目前國際醫藥市場上最重要的藥物主流,然而30年來全世界累積抗體的研究能量,95%抗體仍以老鼠抗體為主,若能夠將其人類化甚至全人類化,將可以有效加速這些抗體藥物進入臨床應用。傳統進行抗體人類化方式,抗體往往會大大降低活性甚至失去功能,若能配合電腦結構模擬輔助抗體(含全人類化)之基因工程技術,將可依不同鼠源抗體,量身打造接近原抗體親和力之人類化甚至全人類化抗體,徹底改革傳統抗體人類化"有功能就好"的傳統觀念,減低抗體藥物開發的時程與成本,讓更多的抗體藥物可以快速進到臨床應用。

3. 人類B 細胞不死化平台(全人化抗體藥開發)
過去研究發現,癌症組織內會累積腫瘤浸潤B細胞(Tumor-infiltrating B lymphocyte; TIL-B),而這些B細胞所分泌之抗體,對癌症組織具有專一性。將B細胞與細胞膜上表現CD40 ligand (CD40L)的細胞共同培養及送入Bcl-2-like protein 1 (BCL-2L-1)及B-cell lymphoma 6 protein (BCL-6)兩種基因,使B細胞達到不死化(immortalized)的效果,建立各種病患(癌症、白斑、病毒或其他感染者)的不死化B細胞抗體庫,以利診斷抗體及全人化治療性抗體之篩選,一旦篩選到中和性抗體藥物就是全人化抗體,不需人類化可大大加速抗體進任臨床之進程。
